As a proof of principle the project will analyse the effect of knocking down all Wnt genes in three compartments of the developing kidney on three different Wnt controlled pathways. If successful, this experiment will not only demonstrate the effectiveness of the system, but also provide valuable information on the complexities of Wnt signalling pathways during development.

Dr Peter Hohenstein has received a £600.000- grant from the NC3Rs (National Centre for the Replacement, Refinement and Reduction of Animals in Research) for a three year project called 'Reducing mouse numbers in complex genetic experiments'.
In this project all the latest technology for the generation of over-expression, knock-down and reporter constructs, targeted integration of these constructs into the mouse genome in ES cells, the generation of fully ES derived ('F0') embryos and mice and time-lapse imaging-based phenotyping will be optimized and combined into one flexible and integrated vector/ES cell system. Especially for experiments where several mutations or other alleles (like Cre expression or reporter alleles) are to be combined, this system can lead to a reduction in animal numbers of up to 90%, as well as reductions in costs and time.
